Schistosoma mansoni is a helminth that causes schistosomiasis in humans. It is one of the so-called neglected tropical diseases. S. mansoni cercariae, which are infective to humans, live in freshwater. They survive in the water by metabolising their glycogen stores which they mainly break down to CO2. Upon entry of the human host, S. mansoni cercariae encounter high glucose concentrations. When subjected to high glucose, S. mansoni cercariae suddenly starts to make lactate – in addition to CO2. Part of the lactate production pathway in S. mansoni is an FBP-activatable lactate dehydrogenase, but its role in metabolism is poorly understood.

In this project, you will make and use a kinetic model to identify possible roles of this allosteric activation. You will fuel a metabolic model of yeast glycolysis with the available kinetic data on the LDH and some of the other glycolytic enzymes of S. mansoni and then interrogate this model for the effect of activating LDH. You will also test the effect of uncertainties in our the measured parameter values.
